Contraindications Aciclovir is contraindicated in patients known to be hypersensitive to the drug. Special Precautions Aciclovir should be administered with caution to patients with renal impairment and doses should be adjusted according to creatinine clearance. The risk of renal impairment is increased by the concomitant use of other nephrotoxic drugs. Use In Pregnancy & Lactation Aciclovir is distributed into breast milk following oral administration. A dose of 200 mg five times daily could expose a suckling infant to 300 mcg per kg body-weight daily; caution is therefore recommended when treating breastfeeding mothers. Administration of Aciclovir produces embryotoxic or teratogenic effects in rats, rabbits or mice. Limited data are available on the use of Aciclovir during pregnancy. Caution should therefore be exercised by balancing the potential benefit of treatment against any possible hazard. Side Effects / Adverse Reactions Anaphylaxis, fatigue, headache; dizziness, confusion, hallucinations, drowsiness; somnolence; decreases in haematological indices, increase & reduction of WBC & eosinophilia; abdominal pain, diarrhea, vomiting, nausea, gastrodynia, epigastralgia; arthralgia; elevation of total cholesterol, triglyceride, serum K level. Interactions Increased mean half-life & AUC w/ probenecid. Other drugs affecting renal physiology could potentially influence aciclovir pharmacokinetics.
